Storage of Strains

Long term cryopreservation

Yeast and E. coli strains are most safely stored by cryopreservation at -80°C in YES + 15% Glycerol.

  • Grow strains while shaking in ~2-5ml YES medium till late exponential phase
  • Spin down the cells
  • Resuspend cells in YES + 15% Glycerol
  • Immidately place the appropriate box in the -80°C.
  • Document your strains in the strains catalog and in the -80°C document in the lab Dropbox

Medium time as spores

Spore are very stable and can sustain storage at 4°C for prolonged time, up to multiple months.

  • Take the formed spores.
  • Resuspend them in sterile water.
  • Place at 4°C.
  • Haploid and diploid cells will mostly not survive this treatment for more than a few weeks.

Very short in liquid
