Lallzyme is a lytic enzyme that digests the cell wall of fungi. It is commercially available for wine production to digest botrytis.

It can also be used to digest fission yeast cells to generate protoplasts for example for DNA extraction and it can be used for our Kill all but spores protocol.

Even though it does not perform as well as zymolyase or Gluculase per weight enzyme (Flor-Parra et al 2014, doi:10.1002/yea.2994) it still performs much better per euro.

Note that the efficiency of the enzyme mix goes down with time! We noticed that after ~6 months to 2 years (batch dependent) after opening and continuous refrigeration, the powder was no longer functioning. If in doubt do a quick check and if not functioning, trash the old and use a new bottle.


To use for protoplast creation:

  • Dissolve 50mg Lallzyme MMX powder directly to 0.5 ml isotonic solution (e.g. Buffer Y1 or 1.2M Sorbitol)
  • Mix until dissolved
  • Filter sterilize when needed (For DNA extractions no sterilization required)


For Kill all but spores:

  • make a 100 mg/ml MMX in water (check if )
  • filter sterilize
  • can be kept at 4°C for at least 1 week without any noticeable effect.
