Hygromycin

Hygromycin is used a dominant resistance marker that is encoded by hphMX. See Hentges et al. 2005 Yeast.

Make a stock solution of 100mg/ml

  • dissolving 1 g hygromycin in 10ml H2O and filter sterilize.
  • filter sterilize and aliquot in 1ml tubes.

Hygromicin is like all antibiotics stored at -20C in the top drawer of freezer 7 (except for G418).

Usage

Pombe:

Add 1:1000 (i.e. 25µl/plate) to solid medium of ~50-60°C to obtain a final concentration of 100µg/ml.

Note that Hygromycin is not working well with EMM due to interactions with the ammonium nitrate, so best to use YES.


Schizophyllum

Add 1:10.000 (i.e. 2.5µl/plate) to solid SMM of ~50-60°C to obtain a final concentration of 10µg/ml.
