SOC

Super Optimal broth with Catabolite repression, is bacterial culture medium that differs slightly with LB broth.

SOC medium is used to grow competent cells after transformation and before plating, to maximize the efficiency of the transformations.

Ingredient	Amount
ddH2O	100 mL
SOB-Medium (powder in chemical room)	3.07 g
MgCl2 1M (final concentration 10 mM]	1 ml
Glucose 60% (final concentration 20 mM)	599 µl

To make:

• Add the SOB-Medium to the water and shake until the solution is dissolved.
• After sterilizing by autoclaving add 599 µl of the 60% Glucose and also
• add 1 ml of MgCl₂ from the 1M Stock solution ( fridge 3).

If the 1 M Stock of MgCl₂ is empty, dissolve 2,033 g of MgCl₂ in 10 ml ddH₂O and filter the solution sterile.

Aliquot the medium in 1,5 ml Eppendorf tubes and freeze in freezer no. 7
